| Definition: | Mitogen-activated protein kinases (MAPKs), also known as extracellular signal-regulated kinases (ERKs), act as integration points for multiple biochemical signals. This protein phosphorylates and activates the p38 MAPK in response to inflammatory cytokines or environmental stress. As an essential component of the p38-mediated signal transduction pathway, this protein is involved in many cellular processes, such as stress-induced cell cycle arrest, transcription activation, and apoptosis. It catalyzes the concomitant phosphorylation of a threonine and a tyrosine residue exclusively in p38, and is probably activated by dual phosphorylation on Ser207 and Thr211. It possesses a very low rate of autophosphorylation. It has two isoforms, MKK6 and MKK6b (shown here), which are produced by alternative splicing. MKK6 is expressed only in skeletal muscle; MKK6b, in contrast, is found in skeletal muscle, heart, and to a lesser extent, in liver and pancreas. It is strongly activated by UV, anisomycin, and osmotic shock but not by phorbol esters, nerve growth factor (NGF), or epidermal growth factor (EGF). It belongs to the MAPK subfamily of the Ser-Thr family of protein kinases. |
